Alchemilla vulgaris has traditionally been used to treat gynecologic diseases. The anti-inflammatory and vasorelaxant effects of the extract of A. vulgaris (AVE) have been reported in previous studies. This study aimed to investigate the effects of A. vulgaris on PCOS, one of the most common metabolic disorders in adult women of reproductive age, accompanied by inflammation and vascular endothelial dysfunction, in an experimental animal model. Three groups of prepubertal female Wistar rats were established: PCOS, Control, and PCOS + AVE groups. Subcutaneous dehydroepiandrosterone (DHEA) was administered for 35 days to the PCOS and PCOS + AVE groups. The rats in the PCOS + AVE group were orally administered AVE (50 mg/100 g bw/day) for 35 days. Systemic blood pressure, vascular endothelial function, ovarian morphology, and endocrine biomarker levels were measured at the end of the experiment. TNF-α levels were also evaluated in the thoracic aorta and ovaries. Hormonal imbalances, ovarian morphological changes, and endothelial dysfunction were observed in the PCOS group, which returned to the controls after AVE treatment. Furthermore, AVE attenuated the aortic and ovarian levels of TNF-α in the PCOS group. Our study suggests that AVE may have beneficial effects in the treatment of PCOS due to its anti-inflammatory and vasculoprotective properties.

Oztas, Berrin; Eraldemir, Fatma Ceyla; Akbal, Sezgi; Acar, Esra; Hunc, Fatih; Yilmaz, Melda Yardimoglu

Objectives: The aim was to investigate the effects of cinnamon on the kidney tissue serum endoplasmic reticulum (ER) stress marker, reticulone (RTN)1A, receptor for advanced glycation end products (RAGE) and the lipid peroxidation indicator, malondialdehyde (MDA) in an experimental diabetes mellitus (DM) rat model. Methods: Twenty-eight male Wistar Albino rats (six months old and weighing 350–400 g) were divided equally into four groups: 1) Control group-citrate buffer (0.2 M, pH 4.4; ip); 2) Cinnamon group-cinnamon (600 mg/kg/day, orogas-tric tube); 3) DM group-STZ (35 mg/kg, ip); and 4) DM + cinnamon group; Cinnamon and STZ were given at the same doses and route as in Groups 2 and 3, respectively. At the end of the 12-week experiment period, serum, urine and kidney tissue samples were taken from all groups. Serum RTN 1A, RAGE, MDA, urea, blood urea nitrogen (BUN), creatinine levels andkidney tissue RTN 1A, RAGE and MDA levels were measured. Results: Our biochemical results showed that there was a statistically significant decrease in RAGE and MDA levels in the DM + cinnamon group compared to the DM group (p<0.05). In addition, the decrease in serum urea, BUN, and creatinine levels in the DM + cinnamon group was also remarkable (p<0.05). Althought histologically no widespread necrosis was observed, cortical interstitial vascular dilatation was observed in DM+cinnamon group. Conclusion: Cinnamon was effective in reducing markers of oxidative stress and ER stress including RAGE and MDA, in kidney tissue in an animal model of diabetic nephropathy.

Effect of chronic stress on serum and tissue levels of klotho and tnf-α

Acar, Esra; Colak, Tuncay; Eraldemir, Fatma Ceyla; Yener, Mehmet Deniz

Objective: Chronic stress is recognized as a factor that affects almost all organs and tissues and disrupts homeostasis. Stress also directly impacts the inflammatory process. In addition, klotho is a protein associated with lifespan. This study investigates the relationship that chronic stress has with klotho and tumor necrosis factor alpha (TNF-α), which is a key protein in the proinflammatory process. Materials and Methods: The study involves 16 Wistar albino rats divided into control and stress groups. The study applies the protocol for the chronic unpredictable mild stress (CUMS) model and uses the enzymelinked immunosorbent assay (ELISA) method to measure the TNF-α and Klotho levels in the serum, heart, aorta, and liver tissues. Results: The serum samples obtained from the stress group were found to have a significant decrease in klotho levels (p=0.013) and high TNF-α levels (p=0.042). However, no significant difference was observed in the TNF-α or klotho levels in the heart, aorta, and liver tissues between groups. Conclusion: Under chronic stress, a significant increase is observed in TNF- α, one of the main cytokines of inflammation, as well as a decrease in klotho hormone. This research endeavors to contribute to the existing body of knowledge concerning the interplay among stress, inflammation, and longevity.

Intraductal botulinum toxin injection suppressed the inflammation in experimental acute pancreatitis; [Deneysel akut pankreatit modelinde intraduktal botulinum toksin uygulaması enflamasyonu baskıladı]

Yılmaz, Tonguç Utku; Eraldemir, Fatma Ceyla; Gürel, Bora; Yavuz, Ömer; Acar, Esra; İşken, Tonguç; Yılmaz, Hasan; Utkan, Nihat Zafer

BACKGROUND: Acute pancreatitis (AP) is inflammation of pancreas in which pancreas enzymatic activity is increased. Parasym-pathetic innervation of pancreas plays an important role in several functions of pancreas. Botulinum toxin (BTx) might be a tool to suppress the pancreas activity in AP. METHODS: In the preliminary experimental study, BTx (15U/kg) was administered directly and intraductal ways. After 10 days, blood amylase, lipase, trypsinogen, insulin, and glucagon levels were compared and no significant difference was seen between groups. Intraductal BTx administration is preferred for experimental AP model in rats; control, AP, intraductal BTx, and AP with Intraductal BTx (AP+BTx). AP was created by intraperitoneal injection of cerulean 20 µg/kg/injection (5 times). After 24 h, serum amylase, lipase, IL-6, IL-1β, TNF-α, and IL-10 were measured and pancreas tissue was evaluated for inflammation and necrosis. RESULTS: Mean serum amylase, lipase IL-6, IL-1β, and TNF-α levels of the AP group were significantly higher compared to the other groups (p<0.05). However, there was no significant difference between the amylase and lipase levels of control, BTx, and AP+BTx groups. Serum insulin and glucagon levels in AP group were significantly higher than control and BTx groups (p<0.05). However, there is no significant difference between the insulin and glucagon levels of AP and AP+BTx groups. in pathological evaluation. In AP+ BTx group, there is less amount of centrilobular necrosis and there is mild inflammation and hyperplasia of pancreatic duct epithelium. CONCLUSION: Administration of intraductal BTx suppressed the AP without making significant suppression in endogenous activity of pancreas.
