Preservation of septal cartilage for early revisions: a histopathological study

Hancı, Deniz; Kumaş, Ömer; Turgut, Hüseyin; Çelik, Cem; Uygan, Uğur; Altun, Hüseyin; Onaran, Öykü İzel; Gürpınar, Berk; Obuz, Mert; Uyar, Yavuz

Objective: To determine how storage medium and temperature affect the histological viability of autologous septal cartilage intended for early revision rhinoplasty. Methods: Cartilage specimens stored for 6 and 12 months in four media—dry, gentamicin, cefazolin, and saline—at two distinct temperatures underwent histopathological evaluation after paraffin embedding. The assessed parameters were graft viability, fibrosis, osteoid metaplasia, and peripheral chondroid proliferation. Results: Among 480 septal cartilage specimens harvested from 30 patients, overall viability fell from 62 ± 15% at 6 months to 44 ± 14% at 12 months (p < 0.001), while fibrosis, osteoid metaplasia, and peripheral chondroid proliferation all increased significantly (each p < 0.001). Viability declined in every storage condition, but grafts kept at − 80 °C or in saline-based solutions deteriorated fastest, reaching a mean viability of 39% versus 53% for dry storage at − 20 °C at 12 months. Multivariate analysis confirmed − 80 °C storage (OR 2.3, 95% CI 1.4–3.8) and saline media (OR 1.9, 1.1–3.4) as independent predictors of poor viability (< 40%), whereas antibiotic additives were not protective. Histological evidence of osteoid metaplasia (OR 3.6, 1.9–6.8) and peripheral chondroid proliferation (OR 2.1, 1.2–3.7) strongly predicted non-viable grafts. A linear mixed-effects model demonstrated additive negative effects of − 80 °C and saline on both baseline viability and its rate of decline (marginal R² = 0.52). Dry storage at − 20 °C yielded the most favorable long-term viability. Conclusion: Dry storage of autologous septal cartilage at − 20 °C preserves graft viability well enough to offer a cost-effective alternative for early revision surgeries, warranting confirmation in prospective clinical trials.

CRISPR screens reveal YAP/TEAD axis as a mediator of drug-tolerant cells in EGFR-mutant NSCLC

    Yusuf, Muhammed

Despite initial responses, most patients with metastatic lung cancer—including those with EGFR mutations—ultimately develop resistance to targeted therapies. To systematically uncover mechanisms underlying this resistance, genome-wide CRISPR knockout and activation screens were conducted in EGFR-mutant lung cancer cell lines treated with EGFR inhibitors such as osimertinib and gefitinib. These screens highlighted a recurrent involvement of genes associated with the Hippo signaling pathway. Notably, a subset of tumor cells, termed 'persister' cells, survive initial osimertinib exposure by engaging non-genetic, transcriptional adaptation mechanisms that promote drug tolerance. Our studies, integrating both genetic and pharmacological approaches, identified Hippo pathway activation as a key driver of this drug-tolerant state. Importantly, co-inhibition of EGFR and the Hippo signaling axis led to a pronounced reduction in cell viability in both established cell lines and patient-derived organoids. These findings propose that dual targeting of EGFR and Hippo signaling may offer a promising therapeutic approach to overcome resistance in EGFR-mutant lung cancer.

    Meta-analysis of laser-assisted periodontal therapy (2015–2025)

Sunar, Ayşegül; Sır, Eda; Taş, Çağan; Tekin, Enes

This systematic review and meta-analysis evaluated the adjunctive clinical efficacy of laser-assisted periodontal therapy compared with conventional scaling and root planing (SRP). The primary research question was whether laser systems provide additional improvements in clinical attachment level (CAL) and probing depth (PD) in patients with periodontitis. PubMed, Scopus, and Web of Science were searched for studies published between January 2015 and October 2025 following PRISMA 2020 guidelines. Randomized controlled trials and prospective clinical studies comparing laser-assisted periodontal therapy with SRP alone were included. Primary outcomes were changes in CAL and PD with a minimum follow-up of 3 months. Meta-analysis was performed using a random-effects model when sufficient quantitative data were available. Risk of bias was assessed using the Cochrane RoB 2.0 tool, and evidence certainty was evaluated using GRADE. Twenty-six studies were included in the qualitative synthesis, and quantitative meta-analysis was feasible only for Er: YAG lasers. The pooled mean difference in CAL gain favored Er: YAG adjunctive therapy (MD = 0.319 mm; 95% CI: −0.097 to 0.735 mm), without statistical significance. Moderate heterogeneity was observed (I² ≈ 54%). Evidence for diode lasers, Nd: YAG lasers, and antimicrobial photodynamic therapy showed modest short-term effects but lacked standardized data for quantitative pooling. Laser-assisted periodontal therapy may provide small adjunctive benefits over SRP alone. Er: YAG lasers showed the most consistent, though clinically limited, effects and should be considered supplementary rather than routine adjuncts.

A comparative analysis of the utilization of upper and lower extremity functions in mental chronometry tests in individuals with multiple sclerosis

Şekercan, Gizem; Yetkin, Mehmet Fatih; Karabudak, Rana; Tuncer, Aslı; Salcı, Yeliz

Background: Mental chronometry tests, including upper and lower extremity functions, evaluate the temporal congruence component of motor imagery. This study aimed to compare the utilization of upper and lower extremity functions in mental chronometry tests. Methods: This study included 70 individuals with multiple sclerosis and was designed based on an a priori power analysis. Mental chronometry tests, including the Box and Block Test (BBT) and the Timed Up and Go Test (TUG), were administered, and execution times, motor imagery times, and mental chronometry ratios were compared between the most and least affected extremities. Results: A significant difference was observed in execution times between the most and least affected extremities in BBT (p = 0.001). Similarly, motor imagery times in BBT were significantly different between most and least affected extremities (p = 0.009). However, no significant difference was found in mental chronometry ratio between the most and least affected extremities (p = 0.179). Comparisons between upper and lower extremity tests revealed that the mental chronometry ratio was significantly higher for BBT than for TUG test in both the most affected and least affected extremities (p = 0.001). Conclusion: Individuals with MS showed differences in imagery–execution congruence between upper- and lower-extremity mental chronometry tasks, with higher congruence observed in the upper-extremity task. These findings support a task-specific interpretation of motor imagery performance rather than a generalized advantage of upper-extremity motor imagery. Trial registration: Not applicable. This study was observational and did not involve a clinical trial registration.

Preparation of semi-transparent V2AlC thin films using the thermionic vacuum arc technique: crystal and substrate effects on the electrochemical performance

Elmas, Saliha; Rafighi, Mohammad; Mohammadigharehbagh, Reza; Akırtın, Ahmet; Pat, Suat; Korkmaz, Şadan

Among the MAX phases, vanadium aluminum carbide (V2AlC) is one of the remarkable elements due to its superior properties. These intriguing properties stem from physical characteristics such as crystallinity, grain size, and so on, which further affect its electrochemical performance. One of the major and significant issues in this way is using an appropriate substrate to form the desired crystal orientation. According to the results, the crystal nature of the substrate and conductive layer illuminates the formation of polycrystalline V2AlC with various preferential orientations. This subject also directly changed the electrochemical performance, Li-ion diffusion, and resistivity of the V2AlC thin film. The XRD, high-resolution XPS, and Raman spectroscopy results revealed V2AlC formation on all substrates. In all cases, the surface characteristics were confirmed nano-dimensional growth, with a high level of compactness, homogeneity, and a crack-free landscape. Moreover, the optical measurements exhibited semi-transparency of the thin film with lower disorder but higher cutoff energy when coated upon the ITO/PET substrate. The electrochemical results highlighted that the V2AlC coated upon the ITO/PET platform outperforms due to non-Faradaic behavior, lower disorder, and small grain size, facilitating Li-ion diffusion and more capacity storage behavior inside the aqueous LiClO4-PC (0.1 M). Subsequently, the selected material paved the way for using it as a promising candidate in metal-ion storage.

Predictive factors associated with the safety and efficacy of refractive surgery for Myopia: a retrospective cohort study

    Yenihayat, Fatih; Sumer, Fatma

Background/Objectives: Wavefront-guided refractive surgery delivers excellent visual outcomes for myopia, but the relative contribution of demographic, refractive, and corneal preoperative variables to safety and efficacy remains incompletely characterized in real-world single-surgeon cohorts. We aimed to identify independent predictors of safety and efficacy after wavefront-guided femtosecond LASIK (i-LASIK) and wavefront-guided PRK (wfPRK) for myopia, using a methodologically robust analytical approach. Methods: Single-center, single-surgeon retrospective cohort of consecutive myopic patients operated between January 2020 and July 2024. After exclusion of hyperopia, ectatic disorders, incomplete follow-up, and amblyopia (preoperative best-corrected visual acuity [BCVA] logMAR ≥ 0.3), 329 patients (648 eyes) were analyzed. The primary outcomes were the safety index (postoperative BCVA / preoperative BCVA) and the efficacy index (postoperative uncorrected visual acuity / preoperative BCVA), modelled as continuous variables. To preserve the independence assumption while making full use of bilateral data, all inferential analyses — Spearman correlations, Mann–Whitney U / Kruskal–Wallis H tests, and multivariable linear regression with eight preoperative predictors entered simultaneously — were performed separately for right and left eyes. Standardized regression coefficients (β) and 95% confidence intervals are reported. Results: The cohort included 162 male (49.2%) and 167 female (50.8%) patients, mean age 30.0 ± 6.4 years; 411 eyes (63.4%) underwent i-LASIK and 237 (36.6%) wfPRK. Median follow-up was 101 days (range 23 days to 4.2 years). Overall, 99.4% of eyes had a safety index above 0.85 and 98.8% had an efficacy index above 0.80. Across all four multivariable models (safety and efficacy, right and left eyes), preoperative BCVA logMAR was the strongest independent predictor (β = +0.437 to +0.682, all p < 0.001). Preoperative astigmatism was independently associated with reduced left eye safety (β = −0.102, p = 0.028), and high myopia (SEQ ≤ −6.01 D) with reduced right eye safety (β = −0.096, p = 0.040) and efficacy (β = −0.113, p = 0.043). Age, sex, and surgical technique (i-LASIK vs wfPRK) were not independently associated with either index (all p > 0.05). Conclusions: In a real-world wavefront-guided refractive surgery cohort, both i-LASIK and wfPRK delivered comparably high safety and efficacy. The strongest determinant of both indices was preoperative BCVA logMAR, while preoperative astigmatism and high myopia showed small but consistent independent contributions. The absence of an independent age, sex, or technique effect supports an indication-based rather than procedure-based decision framework and a more nuanced view of refractive surgery outcome prediction.

Synthesis and Characterization of Iron-Cobalt@Activated Carbon Nanoparticles for Electrochemical Sensing of Dopamine Neurotransmitters

Altuner, Elif Esra; Janabi, Muhammad Yousif Ali Al; Sen, Fatih; Dar, Umar Ali

Activated carbon-supported FeCo@AC nanoparticles (FeCo@ACNPs) were synthesized for catalytic activity and extend shelf life by incorporating bimetallic iron-cobalt nanoparticles (FeCoNPs). The synthesized nanoparticles were characterized using fourier-transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), and X-ray diffraction (XRD) to elucidate their structural and morphological properties. FTIR analysis confirmed the presence of metal nanoparticles with characteristic peaks at 590–612 cm⁻¹. TEM analysis revealed an average nanoparticle size of 10.71 ± 3.52 nm, while XRD results indicated crystalline dimensions of 7.2 nm. FeCo@ACNPs were further modified onto a glassy carbon electrode, and their sensor activity was evaluated for dopamine detection in the linear range of 0.1–1 mM. The study determined a limit of detection (LOD) of 0.65 µM and a limit of quantification (LOQ) of 1.97 nM. These findings not only reinforce previous studies on dopamine sensing but also provide valuable understanding of dopamine sensing and highlight its potential for health applications, particularly in diagnosing neurological disorders like Parkinson’s disease by monitoring dopamine levels.

Comparison of surgical approaches to the hippocampal formation with artificial intelligence

Dundar, Tolga Turan; Pehlivanoğlu, Meltem Kurt; Eker, Ayşe Gül; Albayrak, Nur Banu; Mutluer, Ahmet Serdar; Yurtsever, İsmail; Doğan, İhsan; Duru, Nevcihan; Türe, Uğur

The relatively complex functional anatomy of the mediobasal temporal region makes surgical approaches to this area challenging. Several studies describe various surgical approaches, along with their combinations and modifications, to reach lesions of this region. Some of these surgical approaches have been compared using artificial intelligence-based approaches that can be predicted, classified, and analyzed for complex data. Several surgical approaches, such as anterior transsylvian, trans-superior temporal sulcus, trans-middle temporal gyrus, subtemporal–transparahippocampal, presigmoid-retrolabyrinthine, supratentorial-infraoccipital, and paramedian supracerebellar-transtentorial, were selected for comparison. Magnetic resonance images (MRIs) were taken according to the criteria specified by the Radiology Department. With an open-source software tool, volumetric data from cranial MRIs were segmented and anatomical structures in the main regions were reconstructed. The Q-learning algorithm was used to find pathways similar to these standard surgical pathways. The Q-learning scores among the selected pathways are as follows: anterior transsylvian (Q_A) = 31.01, trans-superior temporal sulcus (Q_B) = 25.00, trans-middle temporal gyrus (Q_C) = 28.92, subtemporal-transparahippocampal (Q_D) = 23.51, presigmoid- retrolabyrinthine (Q_E) = 27.54, supratentorial-infraoccipital (Q_F) = 27.2, and paramedian supracerebellar-transtentorial (Q _G) = 21.04. The Q-value score for the supracerebellar transtentorial approach was the highest among the examined approaches and therefore optimal. A difference was also found between the total risk score of all points with pathways drawn by clinicians and the total risk scores of the pathways formed and followed by Q-learning. Artificial intelligence-based approaches may significantly contribute to the success of the surgical approaches examined. Furthermore, artificial intelligence can contribute to clinical outcomes in both preoperative surgical planning and intraoperative technical equipment-assisted neurosurgery. However, further studies with more detailed data are needed for more sensitive results.

Subcortical volume alterations indicate structural atrophy patterns over time in relapsing–remitting multiple sclerosis: a volBrain-based analysis

Kılıç, Buket; Tekin, Ayla; Bünül, Sena Destan; Efendi, Hüsnü; Çakır, Özgür; Kılıç, Kamil Can; Çolak, Tuncay

Background: Multiple Sclerosis (MS), a neurodegenerative disorder affecting central nervous system, predominantly manifests as Relapsing–Remitting MS (RRMS), with subcortical volume loss serving as a prognostic indicator. Aims: This study aimed to assess time-dependent volumetric changes in thalamus and basal nuclei using a validated automated tool, volBrain software, on MR images (MRI) across three years and relationship between structural changes and disease progression. Methods: The study included 50 RRMS patients (33.5 ± 6.3 years; 68% female, 32% male) and 50 healthy controls (38.0 ± 5.8 years; 64% female, 36% male). T1-weighted brain MRI scans (from 2017, 2019, 2022) were analyzed via volBrain. Statistical analyses included Shapiro–Wilk, Mann–Whitney U, Friedman, and Dunn tests. Results: Thalamus, nucleus caudatus (nuc. caudatus), and nucleus lentiformis (nuc. lentiformis) volumes were significantly lower in RRMS (p < 0.05). Significant volume loss was observed between 1st-3rd MRI and 2nd-3rd MRI (p < 0.05), but difference between 1st-2nd MRI was not statistically significant (p > 0.05). The most pronounced volume loss occurred between 1st-3rd MRI, with the greatest reduction observed in left nuc. caudatus (7.92%). This loss was followed by right thalamus (5.68%) and nuc. lentiformis (4.35%). Although atrophy was observed across disease duration groups, this difference was not statistically significant (p > 0.05). Volume loss in thalamus and basal nuclei indicated significant atrophy in RRMS patients. Conclusions: Findings highlighted MS-induced subcortical atrophy that significantly impacts brain structure. This study was distinguished through volBrain, highlighting its effectiveness in providing data with clinical relevance. Subcortical volume changes show potential as biomarkers for disease progression and guiding targeted therapies.

xT-STRIDE threat model for unmanned air vehicle security

Yerden, Aytac Ugur; Senol, Sinan; Kara, Mehmet; Dilibal, Savas

In recent years, substantial progress has been made in the design and development of unmanned aerial vehicles (UAVs) with diverse functional capabilities for avionic applications. Beyond addressing performance bottlenecks, the growing threat of cybersecurity attacks introduces additional complexity to the UAV design process. Developing a high-performance UAV system involves navigating technical security challenges that must be addressed pragmatically. Implementing excessive security measures can increase costs and degrade system performance, while insufficient protection of vulnerable components may expose the UAV to potential exploitation. This study introduces a multi-layered drone system, thoroughly examines threats to its components, and outlines corresponding safeguards in trusted layers. The security requirements for UAV systems were defined using the Common Criteria framework. To strengthen UAV security under real-world operational conditions, a novel threat modeling approach, xT-STRIDE, has been developed. This method incorporates the UAV’s communication and embedded system architecture, modeled with realistic security elements. The xT-STRIDE framework was evaluated theoretically in conjunction with traditional threat modeling techniques.

    Eminov, Elmin; Eminov, Ayşe

Introduction: According to the World Health Organization, adolescent pregnancy is defined as pregnancies of women aged 19 and below. The study aims to analyze the rates of adolescent pregnancies and maternal and fetal outcomes among births within the hospital and compare them with adult pregnancies. Methods: The study is conducted retrospectively in one of Turkey’s socio-economically underdeveloped provinces. The study comprises 16,985 women: 1719 adolescents and 15,266 adults who gave birth in the hospital between January 1, 2020, and December 31, 2023. All data were recorded in the SPSS 28.0 program, and the Kolmogorov–Smirnov test, Chi-Square test, ANOVA, and Independent Simple T-test were applied to analyze the data. Results: In the study, the adolescent pregnancy rate is found to be 10,1%. The mean maternal age (p = 0.000), gravida (p = 0.000), parity (p = 0.000), and number of abortions (p = 0.002) are significantly higher in the adult group. No difference is found between the groups in terms of gestational age (p = 0.067). Newborn birth weight was significantly higher in the adult group (p = 0.000). Cesarean section rates are higher in the adult group (p = 0.001). No difference is found in terms of stillbirth rates. No difference is found between the groups in terms of pre-eclampsia (p = 0.792). No difference is found between the groups in terms of preterm birth (p = 0.664). Conclusion: In conclusion, it came out that, according to the results of the study, the rates of premature birth, pre-eclampsia, and stillbirth in adolescents and the first and fifth-minute Apgar scores are similar to adults. However, newborn birth weights are lower in the babies of adolescent pregnant women. In addition, cesarean section rates are higher in the adult group.

An Ordered Flow Shop Scheduling Problem

Çakmak, Aslıhan; Ceylan, Zeynep; Bulkan, Serol; Şen, Z.; Uygun, O.; Erden, C.

The subject of the study is ordered flow shop scheduling problems which were first seen in the literature in the 1970s. The main objective is to get a fast and good solution. For the purpose of the study, firstly, the ordered flow shop scheduling problems were defined. Then, a heuristic method was suggested for the ordered flow shop scheduling problems and a sample problem was solved and discussed. A genetic algorithm (GA) based on the complexity property was developed. Full enumeration was applied to determine the optimal makespan values based on Smith’s rule because Smith had specified the conditions under which circumstances the permutation can be optimum. This study is one of the few studies in the literature to obtain optimum solutions up to 15 jobs ordered flow shop problems in a very short amount of time. The developed GA heuristic can also solve large size ordered flow shop scheduling problems very fast. The significant advantage of the proposed method is that, while Smith’s method rule not does not work with large size problems since it requires full enumeration to identify best solution to follow a convexity property where GA algorithm can find a good solution very fast.

Comparison of the effectiveness of dry needling and high-intensity laser therapy in the treatment of myofascial pain syndrome: a randomized single-blind controlled study

    Ahi, Emine Dundar; Sirzai, Hulya

Myofascial pain syndrome (MPS) is a very common disease in the population that seriously affects quality of life. Although many treatment modalities are used, there is still no common protocol. The aim of this study was to compare the effectiveness of high-intensity laser therapy (HILT) and dry needling options. This prospective study included 108 patients with neck and/or upper back pain, diagnosed with MPS, who were randomly separated into 3 groups: the exercise group, the exercise + HILT group (HILT group), and the exercise + dry needling group (needling group). The visual analog scale (VAS), neck disability index (NDI), short form-36 (SF-36) scores, and neck range of motion (ROM) values of the patients before and after treatment were recorded and compared between the groups. In all 3 groups, the VAS and NDI scores decreased and ROM levels increased after treatment. The results in the HILT and needling groups were statistically significantly better than those of the exercise group (p < 0.05).The addition of HILT and dry needling to exercises is seen as a more successful treatment option to reduce pain in MPS. Clinical trial registration number: NCT05078333.
